The small nuclear 7SK RNA negatively controls transcription by inactivating positive transcription elongation factor b (P-TEFb). We show that 7SK RNA also directly controls the activity of the transcription regulator HMGA1.

RNA localization is a widely utilized strategy employed by cells to spatially restrict protein function. In budding yeast, localized mRNAs form RNA-protein (RNP) complexes with the RNA-binding protein She2p that subsequently associate with the myosin motor protein Myo4p via the adaptor protein She3p (1). The Myo4p/She3p complex is also involved in inheritance of cortical endoplasmic reticulum (cER). To test for a functional link between mRNA localization and cER inheritance aux1 mutants where created since AUX1 is required for cER inheritance. However, although Aux1p localizes to ER membranes, its molecular role in ER inheritance remains elusive.(2)

First, we would like to establish a novel mRNP purification method from Saccharomyces cerevisiae and second, employ this method to study how the absence of RNA binding proteins such as RNA export factors influences mRNP formation. Our work should help to better understand the relationships between mRNP composition and function.

Borna disease virus (BDV) is the causative agent for Borna disease, a non-cytolytic, persistent infection of the central nervous system detected originally among horses in Borna, Germany. The matrix protein of BDV (BDVM), a 16.2 kDa protein that forms homotetramers and octamers, is associated with virus assembly and budding and may also be associated with the regulation of the viral ribonucleoprotein activity. We have recently shown that BDVM binds single stranded RNA. Mutations in BDVM designed to abrogate specific RNA binding exhibit altered oligomerisation and RNA binding behaviour, and elicit potent growth attenuation in cell-based assays.
